DEVELOPMENT, CHARACTERIZATION AND EVALUATION OF A NEW SERIES OF WATER SOLUBLE MOLECULAR WEIGHT GLUCAN STANDARDS
Although high performance size exclusion chromatography (SEC) is usually equipped with laser light scattering (LLS) in academic labs for measurement of molecular weight (Mw) of polymers directly, SEC is yet frequently used in industries and some labs for determination of Mw and Mw distribution, for calibration and evaluation of instruments used in the quality control of synthesis/ processing, and comparing all kinds of measurement methods, because of its efficiency with aspects of time and cost. Since the retention volume in SEC depends on the hydrodynamic volume rather than Mw, the use of commercially available Mw standards such as pullulan and dextran can lead to a significant overestimation of some functional and medicinal polysaccharide having more expanded or rigid chain conformation in solution. This addresses needs to develop new Mw standard that is comparable to pullulan with regards to narrow Mw distribution but invariant chemical structure, thus, a more accurate Mw determination can be achieved. It has received much attention that biological property of polysaccharides appears to be largely dependent on accurate determination of Mw.
