Effect on Markers of Ozidative Damage and Energy Metabolism in Blood Induced by TDI
Toluene diisocyanate was a kind of indoor air pollutant with low toxic, but the most toxic in all diisocyanate esters, which was recognized as a kind of industry allergen. To study the effect on markers of oxidative damage and energy metabolism in blood induced by toluene diisocyanate(TDI). The mice contacted with TDI by respiratory tract in 50L static inhalation tank. The Exposure sustained two weeks, 4 hours per day. The test had three exposed dose groups: 1/2 LC50、1/4 LC50 、1/8 LC50, and one control group(normal feed). The effect of TDI on contents of superoxide dismutase (SOD), malondialdehyde (MDA), and glutathione (GSH) in serum was observed as a measurement for damage due to oxidation. After exposured to TDI, activities of succinate dehydrogenase (SDH), lactate dehydrogenase (LDH) and ATPase in serum were determined with spectrophotometry. The content of MDA has increased significantly, while the activity of SOD and the content of GSH have declined to different degrees. SDH activity of different group has increased significantly too. LDH activity of male mice has increased significantly, while only that of 1/2LC50 female group shows the increase. There has notable fall in activities of Ca2+-Mg2+-ATPase and Na+-K+-ATPase of different group. As a result, TDI exerts the changes in target systems of oxidative damage and energy metabolism through mechanisms of oxidative damage and its interference on energy metabolism, those target systems would be the referenced ones of forepart diagnoses for groups vocationally exposured to TDI.
