Solid-Phase Synthesis of N-Linked Glycans
1.Introduction Asparagine-linked oligosaccharides (N-glycans) are the most prominent in terms of diversity and complexity among the various types of oligosaccharides. They are involved in a variety of important physiological events, such as cell-cell recognition, adhesion, signal transduction, quality control, and circulatory residence of proteins, lthough synthesis of N-glycans has been investigated to evaluate their biological function, an efficient approach to these complex oligosaccharides has yet to be established in terms of (i) selectivity in the glycosyl bond formations,i. e.,-mannosylation and -sialylation, and (ii) a non-tedious purification process during each step of glycosylation and deprotection.
