Changes of Physiology and Biochemistry in Adventitious Root Formation of Castanea moilissima cv. Yanshanhong in Vitro

To analyze the physiological and biochemical changes in adventitious root formation, endogenous hormone content and enzyme activity were measured. Shoot tips of C. mollissima were chosen in vitro to be rapidly micropropagated, inducted to root by two-step method. Shoots were treated with 3.0 mg·L-1 IBA in experimental group while some others without IBA in control group, and the samples were regularly collected and separately measured. The results were as follows: (1) During adventitious root formation, IAA content reduced slightly in the first 3 days, thereafter it rapidly climbed to the peak on the 7th day, and then reduced. ABA content kept steady and less than that of the control groups. CTK and GA3 content descended, and the content of CTK was significantly lower than that of the controls. The curve of the ratio of IAA/ABA was similar with IAA contents, while the ratio of IAA/CTKs curve showed no clear regularity. (2) The activities of IAAO and POD gradually increased in the first 3 days and then descended to minimum on the 7th day, on the contrary, the activity of PPO climbed and reached the peak on the 7th day. (3) The ratio of IAA/ABA and the activity of PPO were the key factors of adventitious root formation of C. mollissima.
