Segregation of SSR and AFLP Loci in Chestnut F1 Hybrids (Castanea sativa×C.crenata/C.mollissima)

Two interspecific chestnut families segregating for resistance traits against ink and blight diseases were characterized at 10 microsatellite (SSR) loci and 2 AFLP combination-generated loci. Inheritance analysis showed that in the C sativa×C. mollissima (SM) progeny all SSR loci segregated as expected, but that in the C. Sativa×C crenata (SC) progeny significant deviations from the expected segregation ratios were found for two SSR loci (EmCs15 and QpZAG15) located in the same linkage group. On the other hand, a preliminary analysis to assess the similarity/dissimilarity between individuals, based on the available AFLP data for the progenies, respective parents and standards for susceptibility and resistance, revealed a close proximity between the mother tree and the susceptible clone and an evenly distribution of the tolerant clones close to certain groups of the offspring. Moreover, some hybrid individuals seemed to have specific AFLPbased genotypes distinguishing them from the rest of the progeny. Although screening of a much higher number of SSR and AFLP loci is needed in order to identify markers linked to resistance phenotypic traits through association studies, these results show already important putative relationships and distinctive variations are worth exploring. We expect that future screening of the individual susceptible/tolerant phenotypes within the progenies will allow us to verify if any of the genotypic differences found so far can be potentially associated with disease resistance.
