Molecular Characterization of Hybrid Castanea Clones for Forest Reproductive Material Certification

TRAGSA applied RAPDs methodology to molecular characterize the Castanea hybrids clone collection that had been on propagation in its own nursery for years. Nowadays, basic materials of all forest Galician nurseries are being tested using this same method for its approval or refusement. Three different 10-mer primers (number 2 and 4 from kit A and number 11 from kit S, Operon Technologies) are necessary to properly identify the whole collection. The system is repetitive, and shows all the clones are different and present their own DNA profile. Therefore, this method is useful for the molecular characterization and certification of the new hybrid Castanea clones included in the Spanish Catalogue. At the same time, four SSRs markers have been tested with the same purpose, due to their high reproducibility and discrimination power.
