Selection of Castanea sativa Mill. for Resistance to Phytophthora cinnamomi: Testing of Selected Clones

An intensive selection of resistant trees of Castanea sativa has been conducted in Galicia (NW of Spain). The selected trees have to be tested for resistance as soon as possible to save resources and accelerate the availability of resistant fores! Reproductive material. So, the excised stem test was conducted over adult stems coming from the field and correlation between this test and substrate inoculation test were studied. Also, performance of adult and rejuvenated material in the excised stem test was compared. Disease progress on adult excised stem is well correlated with the spreading of the disease when inoculating the substrate, so this variable can be used to pre classify the clones. However, correlations between both tests are much higher when employing rejuvenated material. A new in vitro test has been developed, so life expectancy of in vitro explants can be correlated with the progress of the disease in the substrate inoculation test.
