Study of psychrotrophses application to improve bio-treatment efficiency of nitrogen in constructed wetland in cold area during winter
Based on exploratory experiments, three psychrotrophs mediums are determined to culture bacilli, actinomyces and mildew respectively. Cultured psychrotrophses are separated and purified under at the temperature of 6℃. Physiological characteristics of the psychrotrophses are identified and their nitration abilities are inspected by considering the effects of temperature, dissolved oxygen, carbon concentration, bio-retention time and pH. The configurations of the three psychrotrophses are clear and lubricous under microscope. The bacterium is conglobate and Gram-positive, and the actinomyces is filaceous and Gram-negative, while the mildew is bacilliform and Gram-negative. Some psychrotrophses keep dividing and have certain exogenous metabolism abilities under low temperature. A best growth of the psychrotropphses was detected when pH value was between 7 and 8. Biology fluorescence chromatography spectrum tests indicate that all the psychrotrophses present the maximal activity under 15℃, and they still have definite activity under 0℃, but they will lose normal metabolizing abilities when the temperature is above 35℃.
