Establishment of ELISA determination method and detection of bisphenol A in barreled drinking water
Bisphenol A (BPA) is an estrogenic substance which could be found in the environment and do harm to human and animal. In order to build ELISA determination method and discover if Bisphenol A passed into barreled drinking water from barrel. The prepared artifical antigen was used to immune and coat. Antiserume was obtained by immunizing Balb/C mice, and used in an indirect competitive enzyme-linked immunosorbent assay for the determination of bisphenol A. The linear range of bisphenol A calibration curve was 1.0-1.0×106 μg·L-1, the result presents an inhibition concentration at 50% absorbance (IC50) of 0.123 mg·L-1, and the limit of detection (LOD) is 9.934 μg·L-1.The specificity of antiserum was proved well because the across reactivities with benzene, tert-Butylbenzene, hydroquinone and o-hydroxybenzoic acid were found lower than 0.01%, except phenol was 0.26%. The ELISA method also had well recovery. ELASA determination method for monitoring the concentration of BPA the barreled drinking water and found the concentration of BPA at 10th, 20th and 30th was an increasing trend. The deterrmination results of ELISA were consistent with the result of UV spectrophotometry method. So the established ELISA determination method shows specific recognition of bisphenol A and could be applied in detection of environmental bisphenol A. The determination results also confirm BPA will pass into barreled drinking water from the barrel of polycarbonate.
