NEW DEVELOPMENT OF LOW-COST AND HIGH-THROUGHPUT DNA SEQUENCING

The newly developed low-cost and high-throughput DNA sequencers are based on the microarray chip platforms on which genomic DNA (gDNA) fragments fixed arbitrarily on the solid substrates and formed as an random gDNA microarray, and high resolution images of fluourescence or chemical illumination were taken from the gDNA microarray during the nucleotide extension process according the gDNA templates. Here, we are now developing a new sequencing route for next generation DNA sequencing. In our sequencing route, the sequenced DNA template array is fabricated by arbitrarily immobilizing single long DNA molecules containing multi-copies of gDNA fragments, and a new strategy of DNA sequencing called as alpha-thiosequencing, has been developed. The sequencing route shows potential to achieve homogeneous amplification in human genome-wide region during preparation of sequenced templates, and high accuracy in high-throughput DNA sequencing.
