HOMOGENEOUS DETECTION OF DNA SEQUENCES BASED ON MASS SPECTROMETRY IN COMBINATION WITH NANOPARTICLES PROBE

Mass spectrometry (MS) based methodology offers simple, fast and sensitive diagnosis. While it has become the predominate approach in bio-molecular analysis, it has not been suitable for analyzing nucleic acid due to its low ionization efficiency. We have developed an assay of MS in combination with a nanoparticle probe (NP) encoded with reporter mass molecules and successfully employed it in DNA analysis. This assay demonstrated high MS sensitivity, with the ability to detect target DNA in low femtomolar (10-15M) levels, almost four orders of magnitude better than previously reported. This inaugural effort using combined techniques showed an extraordinary analytical capability for differentiating the single nucleotide polymorphism (SNP), which comprises the most abundant source of genetic variation in the human genome. We have also demonstrated that the method can resolve multiple DNA sequence variations with the same mass, which the classic MS methodology is inherently unable to be differentiated.
