DESIGN AND PREPARATION OF MONOLAYER-BARCODED NANOPARTICLES PROBE FOR DNA DETECTION
Mass spectrometry (MS) is one of the rapid developed analytical technologies for bio-molecular in recent years. We have developed monolayer-barcoded nanoparticles (MBN) probe for on-chip DNA hybridization assay utilize laser desorption/ionization time-of-flight Mass spectrometry (LDI-TOF MS) (Angeω Chem Int Ed 2008, 47, 5009 -5012), and it was successfully employed in DNA analysis. In this article, we described the details how we designed and prepared MBN probe and demonstrated that the mass-tag molecules of MBN probe can be detected with high sensitivity, in one atto mole (10-18mol) through LDI-TOF MS. And MBN probe also showed acute melt points when mixed with pure oligonucleotide-modified nanoparticles in the presence of target strands which indicate that MBN probe could be used in diagnosing genetic diseases that contain single nucleotide polymorphism (SNP).
