A GRAVITY-ACTUATED TECHNIQUE FOR DROPLET MICROFLUIDICS

A gravity-actuated technique for on-demand microfluidic sampling, droplet generation, fusion, mixing and staying is first described. T-junction microchannel is designed and standard PDMS fabrication techniques are used to fabricate microchips. Both oil-phase and water-phase are driven only by controlling height difference of oil-phase with a turntable. Droplets of different size and spacing are easily obtained according to experiment requirement. Moreover two droplets with different size and composition are manipulated to perform high-efficient generation, fusion, mixing and staying to assay. The whole process doesnt need any syringe pump, needle, mechanical valve, electronic component and high-speed CCD. Therefore, this technique is very good to avoid the complicated and expensive chip fabrication and can be easily integrated into portable microdevice for a variety of droplet-based application mainly in biological/chemical assays and high content drug screening.
