DEVELOPMENT OF AN SEMI-AUTOMATED SYSTEM WITH A MAGNETIC NANOPARTICLES SEPARATION DEVICE FOR SNPS DETECTION

We have developed an integrated automation system for genetic analysis and gene manipulation. The system, MagStation- A, is equipped with a magnetic separation unit, a heating block, power supply and a control system. Semi-automation of nucleic acid extraction and purification, the most difficult step in automating genetic analysis and SNPs genotyping manipulation, was realized using magnetic particles (MPs) and solid phase single base extension (spSBE), which we have previously developed for automating the handling of MPs. Using this system, the methylenetetrahydrofolate reductase (MTHFR) gene C677T polymorphism of 48 different samples was semi-automated integrated.
