FLOW SOLID PHASE PYROSEQUENCING REACTION ON CAPILLARY MICROFLUIDIC CHIP
A simple flow solid phase pyrosequencing reaction system was introduced with a microfludic chip consisted of fused silica capillary and permanent magnets. The magnetic beads bearing the template ssDNA were magnetically immobilized in the capillary, and the reaction solutions, including washing buffer and detection mixtures containing the bases, enzymes (polymerase, ATP sulfurylase and luciferase) and substrates (adenosine 5-phosphosulfate and luciferin) were sequentially aspirated by a syringe pump through the EP tubes installed on a turning wheel. The signal was effectively enhanced by bioluminometric assay coupled with modified primer extension reactions (BAMPER) in the single-nucleotide polymorphisms (SNP) assay. SNPs typing of rs8130833 and rsl053315 from human chromosome 21 were demonstrated, and the results were in agreement with the established methods.
