MICROCHIP FREE FLOW ELECTROPHORESIS (μ-FFE) FOR ISO-ELECTRIC FOCUSING (IEF)

A microfluidic chip for free flow electrophoresis was prepared simply by glass or PMMA plates with double tape. The separation channel was isolated from the electrolyte reservoirs by positioned photo-polymerization of conductive gel. Continuous iso-electric focusing (IEF) fractionation of a model protein mixture was demonstrated and collected in different outlet reservoirs, followed by capillary electrophoresis characterization. Monolithic bed was synthesized in situ in the separation channel, and a pH gradient was established with ampholine by applying voltage across the channel, and the ampholine was covalently immobilized locally on the bed as shown by a color indicator. Preliminary result shows that gel and ampholyte free chip FFE IEF fractionation is possible, which is very important for proteomics analysis.
