APPLICATION OF KALMAN FILTER IN THE RESEARCH OF MOLECULAR DYNAMIC PROCESSES IN LIVING CELLS
In this report, we developed a method for tracking the dynamics of molecules in total internal reflection fluorescence (TIRF) images of living cells. After segmenting the moving fluorescence points from the original TIRF images by the background subtraction, a target tracking algorithm based on Kalman filter was realized by the forecast function to predict the target region that may arise in the next frame, so that we can proceed to match operator in the smaller estimation range. The algorithm overcomes the shortcomings of the global search. The results showed that the present tracking algorithm can successfully trace a moving point from a large number of TIRF images, and predict the location and its trajectory of the moving fluorescence points accurately. The present imaging processing methods is useful in the study of dynamics of molecules by TIRF and confocal laser scanning microscopy.
