APPLICATION OF REAL-TIME QUANTITATIVE PCR TO ASSESS ENVIRONMENTAL ABUNDANCE OF TOXIC MCROCCYSTIS IN TAIHU LAKE

Environmental abundance of toxic Microcystis in Taihu Lake with real-time quantitative PCR (RTQ-PCR) was studied in this paper. The standard curves of Cyanobacteria, Microcystis and toxic Microcystis were established by RTQ-PCR, respectively. The environmental abundance of Microcystis and toxic Microcystis was demonstrated by Ct ratio of Microcystis and Cyanobacteria and Ct ratio of toxic Microcystis and Cyanobacteria, respectively. The results showed that the ranges of Cyanobacteria, Microcystis and toxic Microcystis were 106 -1011gene copies/μL (R2=0. 994 1) , 104 -109gene copies/μL (R2=0. 993 6) and 102 -105gene copies/μL (R2=0. 995 1) , respectively. During May, June and July of 2009, Algal density in Taihu Lake was 1. 5 × 105, 2. 9 × 105, 1. 8 × 106 cells/mL. The environmental abundance of Microcystis was about 90% and the abundance of toxic Microcystis was up to over 40% in the lake. It is proved that RTQ-PCR is a useful tool for the rapid quantification of environmental abundance of Cyanobacteria, Microcystis and toxic Microcystis and the main Microcystis of Taihu Lake bloom was toxic Microcystis.
