COMPLETELY STABLE STREPTAVIDIN IMMOBILIZED ON MAGNETIC BEADS IN TERMS OF THERMAL TREATMENT CYCLES FOR ADVANCED GENOME SEQUENCING
In the present research, we succeeded to improve the heat resistant property of streptavidin immobilized on magnetic beads (SA - MB) by addition of specific additives into buffer solution. The reactivity of SA - MB was investigated in terms of the hybridization efficiency of complementary single stranded DNA (complementary-ssDNA) with biotinylated single stranded DNA (biotin-ssDNA) immobilized on SA- MB surface. The addition of ammonium sulfate into the buffer solution drastically improved the heat resistant property of SA - MB after heat-treatment at 95°C for 5 min. The hybridization reactivity was retained by the addition of ammonium sulfate even after three cycles of heat-treatment.
