FISHING OUT THE PARTIALLY FOLDED INTERMEDIATES IN GUANIDINE HYDROCHLORIDE INDUCED UNFOLDING-REFOLDING PATHWAY OF TRYPSIN
GdmCl (guanidine hydrochloride)induced unfolding and refolding of porcine pancreas trypsin have been studied by various physicochemical methods including near and far UV CD (Circular dichroism) spectrum, intrinsic fluorescence spectroscopy (intensity, emission maximum wavelength, tryptophans anisotropy etc. ) as well as ANS (8 - anilino - 1 - naphthalene-sulfonic acid) fluorescence. In our present study, the formation of two unfolding intermediates has been established. They were further confirmed by phase diagram approach. Descriptions of protein unfolding-refolding pathway and the intermediates conformations have also been given.
