RAPID AND VISUAL IDENTIFICATION OF SWINE INFLUENZA H1N1 BY LOOP-MEDIATED ISOTHERMAL AMPLIFICATION

Swine influenza H1N1, which is an acute respiratory disease, is seriously threatening humans health. Nowadays, molecule diagnosis is an effective and accurate way to diagnose H1N1. Here, we developed a method based on loop-mediated isothermal amplification (LAMP) for rapidly and visually identifying swine influenza H1N1 on molecule levels. To perform highly efficient amplification, the concentrations of Mg2+, Bst DNA Polymerase, dNTPs and the ratio of outer and inner primers were optimized. The product of LAMP from the clinical sample was successfully detected by three methods including precipitation, fluorescence and electrophoresis. The results showed that the sensitivity could reach 103 copies. Therefore, our developed LAMP method can be used to rapidly and visually identify swine Influenza H1N1.
