STUDY ON THE ACTION MECHANISMS OF DNA POLYMERASES ON DNA SUBSTRATES USING CATIONIC CONJUGATED POLYMERS

Cationic conjugated polymers (CCPs) are applied to study the function mechanisms of DNA polymerases by using fluorescence resonance energy transfer (FRET) technique. Our preliminary results show that two model DNA polymerases (Klenow fragment of DNA polymerase I and Taq DNA polymerases) have different interaction mechanisms with the DNA substrates and dNTPs, indicating the proposed CCPs-based FRET method may be promising to reveal the functional mechanisms of DNA polymerases.
