Performance testing and analysis of electrophoresis process driven by low voltage on the SOI microchip

In this paper, a new approach for amino acids separation on the electrophoresis chip based low voltage-driven electrophoresis was reported in detail. The insulation of the neighboring arrayed-electrodes was indicated by measuring breakdown voltage, which tested as 260V; by means of current monitoring method, the electroosmotic flow (EOF) of SOI-PDMS hybrid electrophoresis microchip was 3.9×10-4 cm2·V-1·s-1;the range of working voltages which selected by measuring the Volt-Ampere characteristic curve was 100-140·Vcm-1.Using the laser-induced fluorescence detector, a set of low-voltage driven electrophoresis separating and detecting system was finally established. Under the optimized conditions, the mixture of FITC-labeled arginine(Arg) and phenylalanine (Phl)was successfully separated with a resolution of 3.14 on the low-voltage driven electrophoresis microchip.
