Ezpression profiling of transporter genes in swine conceptus trophectoderm cells with arginine supplementation using a swine microarray1
Intrauterine growth retardation (IUGR) is a major concern in domestic animal production. Because nutrient transfer across the placenta is essential for fetal growth and development, as well as a placental transport defect in IUGR, we profiled the expression of transporter genes in swine conceptus trophectoderm cells with arginine (arg) supplementation using a swine microarray in this study. Our results showed that there were 7 (3.6%) and 20 (10.4%) higher expression genes, 41 (21.2%) and 38 (19.7%) lower expression genes, and 145 (75.1%) and 135 (69.9%) no expression genes in 100 and 350 μM arg supplemented groups, respectively. Supplementation of 100 μM arg in the medium up-regulated the expression of sodium/glucose cotransporter, member 9 of solute carrier family 16 (monocarboxylic acid transporters), choline transporter-like protein 2,glycine betaine/L-proline ABC transporter, and amino acid system A transporter, while down-regulated the expression of ABC cobalamin/Fe3+-siderophore transporter, when compared with 350 μM arg group. These regulating activities of arg to transporter genes might be related with its improving the survival of porcine embryos/fetuses and enhancing the nutritional provision to the fetuses for supporting their in utero growth.
