Integrin Signaling in the Uterus: Implications for Animal Health and Nutrition
Integrin receptors have been detected in the endometrium of multiple mammalian species, and the expression of integrins is regulated during the uterine cycle in sheep Johnson et al., 2001, pigs Bowen et al., 1996, mice Aplin et al., 1996, and humans Lessey et al., 1994. The αVβ3 and α4β1 integrins on uterine luminal epithelium (LE) define the window of receptivity to implantation in women Lessey et al., 1994, and trophoblast attachment and outgrowth is integrin-dependent, and occurs on extracellular matrix (ECM) proteins Armant et al., 1986. Secreted phosphoprotein 1 (SPP1 or osteopontin) is an ECM protein that binds integrin receptors and has been linked to establishment and maintenance of pregnancy in sheep, pigs, mice, rabbits, goats and humans Johnson et al., 2003a. Disruption of the Spp1 gene in Spp1-/- and Spp1+/- mice decreases pregnancy rates at mid-gestation, and Spp1 -/- females deliver smaller pups than wild-type dams Weintraub et al., 2004. Mammalian target of rapamycin (mTOR or FRAP1) is an evolutionarily conserved serine/threonine kinase that is considered a “nutrient sensing system central to control of cell growth and proliferation through the regulation of protein synthesis Wullschleger et al., 2006. During embryonic development, some molecules that stimulate mTOR activity also stimulate translation of mRNAs critical to conceptus development, including insulin-like growth factor 2 (IGF2) and selected amino acids Martin and Sutherland, 2001. Recently, SPP1 was shown to induce motility in human trophoblast cells through mTOR/FRAP1 signaling Al-Shami et al., 2005.
