ESR Measurement of Radical Reactive Ozygen Species in Soybean Roots in the Response to Aluminum Stress

5-Diethoxyphosphoryl-5-methyl-l-pyrroline-N-oxide (DEPMPO) was used as spin trapping reagent to identify radical ROS in Al-treated soybean roots. In addition to hydroxyl radical adduct, unidentified signals were observed. As a model study, liposome of phosphatidylcholine derived from soybean was incubated with hydroxyl radical. The ESR spectrum was almost identical to that of the root homogenate, indicating that the unidentified signals may be originated from a carbon-centered radical derived from lipid. ESR spectra of Al-treated root showed a consistently increased signal intensity of the unidentified signals. It can be concluded that Al induces hydroxyl radical, which may be converted to the lipid-derived radical.
