miR-292-5p,let-7g Regulating Protein Kinase C ,Effecting β-casine Ezpression in Murine Mammary Epithelial Cell
The mammary gland has the ability to undergo repeated cycles of tightly regulated postnatal proliferation, differentiation, and apoptosis-mediated regression, providing a model to investigate potential regulators of mammary epithelial growth and differentiation. Protein kinase C forms a central control point mammary gland activated cell-signalling events and regulate a wide of intracellular function including induction of β-casein expression. In this study miR-292-5p,let-7g were inhibited in the mouse mammary epithelial cell. By Western blot analysis of whole cell lysates from purified MEC knock down miR-292-5p or let-7g , PKC protein expression was found upregulating. Bioinformatic predictions suggest that the mouse PKC mRNA 3-untranslated region (3-UTR) contains two miR-292-5p target sites, two let-7g target sites. As well as β-casine expression was change with knock down miR-292-5p or let-7g.Conclusion miR-292-5p , let-7g play a role in mammary cell differentiation and mediates the effects of β-casein expression by target PKC gene.
