Biological activity assays and cellular imaging of anti-human sperm protein 17 immunomagnetic nanoparticles

Sperm protein 17 is a novel cancer-testis antigen and can be candidated as a new target for cancer immunotherapy and molecular diagnosis. The anti-human Sperm protein 17 immunomagnetic nanoparticles were prepared by grafting anti-human Sperm protein 17 antibodies on the surface of chitosan-coated magnetic nanoparticles using the linker of 1-ethyl-3-3-dimethylaminopropyl carbodiimide/N-hydroxysuccinimide. The morphology and properties of the nanoparticles were characterized by transmission electronic microscopy, the conjugation of the antibodies was evaluated by native-polyacrylamide gel electrophoresis, the immunologic activity of the particles was evaluated by enzyme linked immunosorbent assay. A set of in vitro magnetic resonance imaging experiments were performed after incubated the immunomagnetic nanoparticles with human Sperm protein 17 gene transfected ovarian cancer HO-8910 cells. The results showed that we successfully grafted the magnetic nanoparticles with anti-Sperm protein 17 antibody and the IMNPs kept good bioactivity. And the immunomagnetic nanoparticles were targeted successfully to the positive cells, and no obviously nonspecific adsorption was observed.
