Chemical synthesis of follicle-stimulating hormone gene and stable ezpression in CHO cells
Based on amino acids sequences of a subunit and β subunit of follicle-stimulating hormone (FSH), gene sequence was optimized which is easy to be highly expressed in eukaryotic cells, and were chemically synthesized. With genetic engineering techniques, α subunit and β subunit genes were cloned into two eukaryotic cell expression vectors PM902 and pcDNA3.1( + ) respectively, and co-transfected into CHO cell lines for construct stable and high expression cell lines. The expressed human FSH can be applied for therapy of infertility, nurture of zygotes in vitro, preparation of polyclonal antibody and monoclonal antibody to detect human FSH and so on.
