Determination of enzymatic enantioselective resolution of racemic epichlorohydrin by GC
Enantiopure epichlorohydrin (ECH), which is a valuable epoxide intermediate for preparing optically active Pharmaceuticals, can be manufactured by enzymatic catalysis enantioselective resolution from racemic epichlorohydrin. In order to characterize the enzymatic reaction, a method of gas chromatography (GC) was developed for determination of enantiomer of epichlorohydrin. Samples diluted with diethyl ether were isolated by ZKAT-Chiral B capillary column with the split ratio 100:1. The results were detected by FID and calculated by internal standard method. The linear rangers of (S)-ECH and (R)-ECH were all between 0. 011 ~ 0. 12 g/mL, The relative standard deviation (KSD) of (S)-ECH was 1. 08%, and RSD of (R)-ECH was 0. 95%. This method is accurate, simple, rapid and cheap for simultaneous determination of glycerine, chloropropanediol and dichloropropanol.
