Identification of a histidine triad protein family protein from Streptococcus suis 2

A whole genome sequence analysis of the Chinese strain 052YH33 of S. suis 2 enabled us to identification of a novel histidine triad protein gene. The 2.7 kb target DNA fragment was successfully amplified and the recombinant HtpS (histidine triad protein of Streptococcus suis) can be produced in E. coli under the induction of IPTG. A 110 kDa protein was purified by affinity chromatography. Western-blot experiment demonstrated the recombinant protein shares strong specific antigenicity. Flow cytometry analysis demonstrated that the protein can be exposed on the surface of 05ZYH33 strain. Animal protection experiments demonstrated that immunizing with HtpS could protect mice against lethal doses of bacteria which suggested the protein could apply as a vaccine candidate.
