Yeast two-hybrid system based screening of caspase-8 interacting proteins, and co-immunoprecipitation validation
Caspase-8, also called FLICE, is involved in the apoptosis process of eukaryotic cells. Recent research has showed that caspase-8 is implicated in both cell apoptosis and proliferation pathways. In addition, it is also required for mouse embryo development. To fully understand the biological functions of caspase-8 in vivo, we screened caspase-8 interacting proteins from mouse T-cell lymphoma and 7. 5 day embryo cDNA libraries by yeast two-hybrid system and got eighteen positive clones, and co-immunoprecipitation (co-IP) experiments validated that one of these proteins called ACT9, interacted with caspase-8 in human 293T cells.
