Protection of a cytidine deaminase gene gainst tozicity of high dose chemotherapy in mice
Background and Aim:To explore the feasibility of transferring cytidine deaminase (CD) gene into mouse bone marrow cells in order to observe the drug resistance of high dose Ara-C and improve the tolerance of myelosuppression following combination chemotherapy. Methods:1 Separation of vectors and drug resistance genesThe retrovirus vectors containing drug resistance genes SFG-CD (containing human mutant Cytidine Deaminase, mCD) was transduced into amphotropic packaging cell (GP-Am12, Am12), and constructed AM12-SFG-CD. These vectors and cell lines were kindly provided by professor Shicheng Zhao (MSKCC).
