Detecting protein structure flezibility via rigid structure identification

Motivation and backgroundTo detect the flexible region in protein structure is an important step to locate the sequence or regional structures related to the function of protein. Previous researches give us guidance that the stability and flexibility are coexistence in protein structure 1, 2. Conventional approaches use global structure alignment tool to reveal the structure variations, which provide messy information about flexible regions.ApproachesWe investigate in the variant structures of the same sequence, and propose a new method to detect the stable and flexible regions from protein structures. Our assumption is that protein structure contains highly stable regional structure to support its protein function, which can be used to detect other flexible regions of the same protein. Therefore, this proposed approach is to detect stable regions via mining frequent itemset method and local structure encoding scheme for fast search on similar local structure. Furthermore, the flexible part of protein can be detected by identifying rigid structural region of proteins.Materials for this framework are collected highresolution structure variations of protein examined by X-ray crystallization.
