Significance of a Genetic Information Database in Maintaining a Culture Collection of Wood Rot Fungi
Introduction Genetic information describing of organisms has been accumulated and assembled into databases at an astonishing pace; however, information degrading basidiomycetes remains incomplete. An inventory of basic information concerning basidiomycetes including wood rot fungi is increasing due to the emphasis on research of wood preservation and biological resource conversion. On the other hand, identification of basidiomycetes by means of traditional techniques is difficult and sometimes produces erroneous results. Recently, molecular biological methods for the classification and identification of basidiomycetes have been developed and used. The goal of this study is to build a database of the genetic information of basidiomycetes, mainly wood rot fungi for users involved in the field of biological wood degradation. The Deterioration Organisms Laboratory (DOL), RISH institute, Kyoto University maintains a large collection of basidiomycetes, which is valuable for studies of wood deterioration and wood utilization. In the present study, the wood rot fungi. The sequences of the full length of the internal transcribed spacer (ITS) region (ITS I, ITS II, and 5.8S rDNA) of the fungi were analyzed to check species of each strain. In present study, some DOL strains which were not confirmed by homology search according to the ITS sequence were analyzed by comparing the sequences with standard type strains stocked in a culture collection center NBRC. Cluster analysis was also conducted for more to cover little and lack of information in the existing database.
