Cloning and Characterization of Genes Encoding Glycoside Hydrolase Family 5 Enzymes from the Basidiomycete Phanerochaete chrysosporium

Introduction The total genome sequences of the basidiomycete Phanerochaete chrysosporium has been disclosed by the DOE Joint Genome Institute in the United States (http://genom.jgi-psf.org/Phchr1/Phchr1.home.html) and demonstrated the potential as a source to produce various glycoside hydrolases (GHs) acting on lignocellulosic biomass. Most of the enzymes, however, have not been well characterized. Thus, recently, we have decided to clone cDNAs encoding various GHs by using the gene information and to produce the recombinant enzymes as an attempt to improve the enzymatic saccharification process of cellulosic biomass. In this study, we have cloned three cDNAs encoding GH family 5 enzymes from P. chrysosporium and characterized by sequence analysis of them.
