Identification of Anthocyanins in Buckwheat Sprouts and Petals

Four anthocyanins, cyanidin 3-O-galactoside (Cy-ga), cyanidin 3-O-glucoside (Cy-gl), cyanidin 3-O-rutinoside (Cy-gl-rh) and cyanidin 3-O-galactophyranosil-rhamnoside (Cy-ga-rh) were isolated from the sprouts of common buckwheat (Fagopyrum esculentum Moench), separated using HPLC and identified using reversed-phase LC/ESI-MS/MS techniques. In tartary buckwheat (Fagopyrum tataricum Gaertner) sprouts, two anthocyanins (Cy-gl and Cy-gl-rh) were identified. Among 19 common and tartary buckwheat varieties/breeding lines, Hokkai T10 contained the highest amounts of anthocyanins. In addition, dark-grown sprouts of Hokkai T10 could accumulate 0.091 and 2.77 mg g-1 dry wt of Cy-gl and Cy-gl-rh whereas other varieties/breeding lines could accumulate trace amounts of anthocyanins. Given its anthocyanin-rich red cotyledons, Hokkai T10 is a promising line for use as the sprouts of Moyashi type and is consequently strongly recommend as a new type of functional food, rich in dietary anthocyanins. Four anthocyanins, Cy-gl, Cy-gl-rh, cyanidin 3-O-rhamonoside (Cy-rh) and Cy-ga-rh were identified from the petals of red flowered common buckwheat. In every varieties tested, Cy-gl-rh was detected as the major anthocyanins and the next is Cy-gl whereas Cy-rh and Cy-ga-rh were trace or not detectable in white and pink flowered buckwheat. The most part of cyanidin moiety was presented as a proanthocyanidin form (PAs-Cy) in every varieties tested. PAs-Cy contents were higher in white or pink flower than in red flowers.
