Effect of pretreatment on extraction of some intracellular products of Rhodotorula glutinis
Three different pretreatment methods, quenched with liquid nitrogen, ultrasonication and digestion by Snailase, were studied to break the cell wall of Rhodotorula glutiniwas for extracting genome and lipid. The result indicated that all of pretreatment methods were advantageous for extraction of protein, genome and lipid, lntracellular protein can be extracted just by ultrasonic pretreatment. However, genome obtained through being quenched by liquid nitrogen was not as good as by Snailase treatment. It was that demolishing of trituration after being quenched by liquid nitrogen brought to the rupture and smaller quantity of intact genome. Lipid extracted by snailase method was light-coioured and had great content in dry cell.
