Response surface optimization of medium composition for phenylalanine ammonia lyase production by recombinant E. coli
A culture medium for phenylalanine ammonia lyase (PAL) production in E. coli was developed following preliminary studies by means of response surface methodology. Central composite design (CCD) was utilized to optimize the medium constituents and explain the combined effects of four medium constituents, viz., glucose, yeast extract, (NH4)2HPO4 and MgSO4. The optimum values for the tested varibles were: glucose 28.2 g/l, yeast extract 5.01 g/l, (NH4) 2HPO4 7.02 g/l and MgSO4 1.5 g/l. A second-order model equation was suggested and then validated experimentally. The model adequacy was very satisfactory as the coefficient of determination was 0.94. The maxium PAL production was 65.36 U/g which showed overall 7.9-fold enchancement over the original medium.
