Description of Spirochaeta saccharolytica sp. nov., an anaerobic hydrogen-producing bacterium

A novel, obligate anaerobic, mesophilic spirochaea strain HT was isolated from Shengli oilfield in Shandong Province, China. Cells of the strain were spiral shaped with sized of 15-50μm by 0.2-0.38μm and Gram-negative. Growth of the strain was observed between 20℃ and 48℃ (optimum 39℃ ), in 0-2% (w/v) NaCl and between pH 4.6 to 8.5 (optimum pH 5.8-7.0). Strain HTT fermented various pentose, hexose disaccharides and polysaccharides, such as xylan and starch but not cellulose, pyruvate and D-mannitol could also be used as sole carbon source. Amino acids did not serve as substrates for growth. Yeast extract was required for growth. The fermentation products from glucose included ethanol, acetic acid, hydrogen and carbon dioxide. Strain HTT was resistant to penicillin G and rifampicin, but sensitive to tetracycline, neomycin and chloramphenicol. The DNA G+C content of strain HTT was 44.6 mol%. 16S rRNA gene sequence analysis revealed that it was closely related to Spirochaeta caldaria DSM 7334T (similarity 97.7%) and DNA-DNA hybridization value is 65%. However, strain HTT differed from Spirochaeta caldaria DSM 7334T by the discrepancies on fermentative products and utilizable carbohydrates. Based on the phylogenetic analysis and phenotypic characteristics, the novel species Spirochaeta saccharolytica sp. nov., is proposed, with the type strain HTT (=CGMCC1.5081T = JCM15036T).
