HYDROLYTIC ACTIVITY OF ALMOND β-GLUCOSIDASE UPON CELLOOLIGOSACCHARIDES
We have made experimental studies into the enzymatic hydrolysis of cellotriose, cellotetraose and cellopentaose at pH 5.0 within the reaction time range of 0 min to 120 min, the temperature range of 30 ℃ to 70 ℃, the β-glucosidase dosage range of 0.0 mg/ml to 0.4 mg/ml, and substrate concentration range 1.0 mg/ml to 3.5 mg/ml. The results indicate that almond β-glucosidase showed higher activity on short cellooligosaccharide than on longer one. The optimal temperature was determined to be 50 ℃. Higher enzyme dosage could improve the hydrolysis of cellotriose and cellotetraose, and inhibit cellopentaose from degrating. It was also found that an increased substrate concentration resulted in decreases in hydrolysis percentage.
