Effects of Microwave/Alkali Pretreatment on Sugarcane Bagasse Enzymatic Hydrolysis and Accessibility

In this study, a novel microwave-assisted alkali pretreatment was used to pretreat sugarcane bagasse (SCB). Compared to traditional alkali pretreatment, microwave-assisted alkali pretreatment had higher rate of weight loss and could increase the cellulose content and decrease the hemicellulose and lignin content. When pretreated at 700 W for 60min, the SCB had the lowest lignin content of 9.69%, hemicellulose of 11.03 % and the highest cellulose content of 69.97 %. The pretreated samples were hydrolyzed by commercial cellulase and the yield of reducing sugar was measured to evaluate the pretreatment effect. When the pretreatment time was 60min, the microwave-assisted pretreated sample at 700 W had the highest reducing sugar recovery rate of 30.8% after hydrolyzed for 12 h, and the traditional alkali pretreated sample was 23.26 %, while the untreated samples were only 13.63 %. Further results suggested that the rate of protein absorption and enzyme absorption increased with the pretreatment time and power, and was higher than the traditional alkali pretreatment. Thus a higher enzyme accessibility could be obtained by this microwave-assisted alkali pretreatment and was consistent with the increase of reducing sugar yield.
