Isolation and Identification of the Strain KW2 Producing Alkaline Cellulase
A bacterial strain(designated KW2).which has high alkaline cellulase activity,was isolated from compost samples.Cells were Gram-positive,rod-shaped and spore forming.Strain KW2 was identified as a strain of Bacillus subtilis based on phenotypic,physiological and biochemical characteristics,as well as 16S rDNA phylogenetic analysis.The cellulase activity(CMCase)of shaking flask culture has reached 236 U/m1.The characteristics of the enzyme showed that the optimum pH value of the strain was 8 and the optimum temperature was 55℃.1t had both,good alkaline endurance and stable enzymatic activity at pH value 5.0-9.0 and had remarkable heat stability under 65℃.The cellulase produced by KW2 may be candidates for use in detergents,treating textiles.
