Roles for Gq/11-mediated Sigmals in the Muscarinic Regulation of Ciliary Muscle Contraction
Experiments were carried out to examine effects of YM-254890,an inhibitor of Gαsubunit of Gq/11 protein (Gαq/11) on contraction,M3-muscarinic receptor (M3R)-operated non-selective cation channel (NSCC) currents and elevation of intracellular Ca2+ concentration (Ca2+1) induced by carbachol (CCh) in bovine ciliary muscle.We also conducted immunofluorescence microscopy using antibodies specific to M3R,Gαq/11 and members of the canonical transient-receptor-potential (TRPC) family which is attracting wide attention as promising molecularcandidates for receptor-operated NSCCs.We obtained thereby results which strongly indicate involvement of Gq/11-mediated signals in the initial phasic component and the ensuing tonic component of the contraction induced by muscarinic stimulation.
