A Method of Single Nucleotide Discrimination with Inner-Mismatched Base Pair Hairpin Structure Probe

An unlabeled single stranded hairpin structure probe with an inner-mismatched base pair has been designed for single nucleotide discrimination.This method has been used as an alternative to molecular beacon for identifying SNPs.Compared with conventional stem-loop(hairpin)probes,the increased hybridization efficiency and specificity has been demonstrated.A SNP locus was chosen and primers were designed to amplify the specific fragment by asymmetric PCR,probes were designed with this method for detecting the specific A/G spot mutation
