Two SRAP markers linked to sweetpotato stem nematode resistance gene in sweetpotato,Ipomoea batatas(L.)Lam.
Sweetpotato,Ipomoea batatas(L.)Lam:cv Xushu 18 which is highly susceptible to sweetpotato stem nematode(SSN),cv.Xu 78-1 witch is highly resistant to SSN,and their F1 progenies were used as research material for seauence-related amplification polymorphism(SRAP)assay in this study.The disease-resistant and susceptible pools,which included 8 highly disease-resistant and 8 highly disease-susceptible strains,were established in the light of the different sensitivity to SSN among the F1 progenies.By the disease-resistant and disease-susceptible pools,we screened out 77 SRAP primer pairs,which could be amplified polymorphic bands between the resistant and susceptible pools,from 225 primer pairs.Through further screening,4 primer pairs which assumed linking to SSN resistance gene were retained.The 4 primer pairs were identified by the two parents,the resistant and susceptible pools,and 65 resistant strains and 79 susceptible strains picked out in F1 progenies.In result,two SRAP markers Iinked to SSN resistance gene obtained.The 2 markers,named as SP1 and SP2,obtained through SRAP-PCR amplified with the primer pair a5b12 and a9b11,whose genetic distance to SSN resistance gene were 4.8 cM and 4.2 cM,respectively.The 2 markers should be used for cloning diseaseresistant gene and molecular marker auxiliary selection on SSN disease-resistant breeding.
