Molecular cloning and expression profile analysis of F3H cDNA encoding flavonoid 3-hydroxylase from the purple-fleshed sweetpotato,Ipomoea batatas(L.)Lam.

Flavonoid 3’-hydroxylase(F3’H,EC.1.14.13.21) is involved in the biosynthetic pathway of anthocyanin and might be the committed step of this pathway.In this study,a full-length cDNA of F3’H was isolated and characterized from purple-fleshed sweetpotato(Ipomoea batatas(L).Lam.).The fulllength IbF3’H cDNA was 1797 bp in length containing a 1554 bp open reading frame(ORF)encoding 518 amino acids.Comparative and bioinformatics analyses revealed that IbF3H had extensive homology with F3Hs from other plant species.Conserved Domain search identified IbF3H as a cytochrome P450.Three F3H-specific motifs(VVVAAS,GGEK and VDVKG)were conserved in IbF3H.Phylogenetic analysis classified IbF3H into the same subgroup with F3H from I.purpurea,I.tricolor and I.nil.IbF3H was expressed in all tested tissues including fibrous roots,thick roots,storage roots,leaves,petioles and stems.During the stage of storage root formation,IbF3H was expressed most abundantly in storage roots.IbF3H expression was strongly associated with anthocyanin accumulation in five different sweetpotato cultivars,suggesting that F3H gene expression was associated with the activation of anthocyanin biosynthesis.
