Analysis of root-specific promoters in sweetpotato

To develop transgenic sweetpotato with high-level expression of useful components in roots,we have screened a large number of root-specific candidate genes from EST analysis and GenBank database search.Through RT-PCR analysis,four genes with root-specific expression were selected for prometer isolation.IbTIP1 and IbSBEI were preferentially expressed in fibrous roots and storage roots,respectively.IbAQP1 transcript was abundant in all parts of roots.The expression level of sporamin gene was high in stem and all the roots.The promoter sequences were isolated by chromosome DNA walking with a long-distance PCR.The complete sequence of each promoter was determined and fused to β-glucuronidase reporter gene(GUSPIus).Transient expression analysis on transverse storage root sections revealed that IbAQP1 and IbSBEI promoters confer high levels of GUS expression in the storage roots of sweetpotato.We are currently generating promoter-GUS transgenic sweetpotato plants to examine GUS activity in root tissues.
