In vitro selection of salt-tolerant mutants and cloning of candidate salt-tolerant genes in sweetpotato(Ipomoea batatas(L.)Lam.)

A total of 276 plants tolerant to NaCl were obtained using in vitro selection of embryogenic suspension cultures of sweetpotato (Ipomoea batotas(L.)Lam.)cv.Lizixiang and gamma-ray induced mutation.Of them 18 plant lines showed significantly higher in vitro salt tolerance,proline contents and superoxide dismutase(SOD)activity than control plants after in vitro assay for salt tolerance and analyses of proline content and SOD activity.Three of 18 plant lines had significantly better growth and rooting ability than the remaining 15 plant lines and control plants at 171 mM NaCl after water culture assay for salt tolerance in a greenhouse.A forward subtractive cDNA library was constructed by using the mutant obtained and the suppression subtractive hybridization(SSH)technique followed by PCR screening.The 395 positive clones were screened from a total of 1312 clones obtained by SSH.The 376 qualified expressed sequence tags (ESTs)blasted by Blast2Go software.ESTs related to catalase,pyrroline-5-carboxylate reductase,dehydration-responsive family protein of salt tolerance in plants were identified.Pyrroline-5-carboxylate reductas,dehydration-responsive family protein genes were isolated and cloned.
