Hydrogen sulfide promotes root organogenesis in sweetpotato seedlings

In this report,we demonstrate that NaHS,a hydrogen sulfide(H2S)donor and gaseous H2S aqueous solutions promote the adventitious root formation mediated by auxin and nitric oxide(NO) signaling transduction.Application with the H2S donor to seedling cuttings of sweetpotato(Ipomoea batatas)could promote the number of adventitious roots in a dose-dependent manner.It was also verified that H2S or HS-rather than other sulfur-containing components derived from NaHS attribute to the progressive role in adventitious root formation.Rapid increment of endogenous H2S,IAA and NO were sequentially observed in sweetpotato seedling after application with the exogenous H2S donor.Combined with exogenous indole acetic acid(IAA)and its inhibitor N-1-naphthylphthalamic acid(NPA),NO donor SNP and its scavenger 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide(cPTIO),further investigations showed that H2S and IAA or NO induced root formation additively;while the H2S-mediated effect was alleviated by NPA and cPTIO.These results jndicted that H2S induced adventitious root formation probably mediated by the IAA and NO pathway and acted upstream of IAA and NO signaling.
